Muscadine Vines has numerous leaves with big yellow areas on them. We are in north Florida. We have never had issues in the past. Had some aphids earlier this year though.
This looks like one of the symptoms of Grapevine Fanleaf Virus. As it progresses, you will notice more deformation.
Complete removal of your grapes and replacing with an unrelated plant species will be necessary. This is not curable.

I have a male and female muscadine grape vines and they have been producing beautiful for 2 to 3 years. For some reason my female grape vine died. What do I for the male? I've tried to buy a female but cannot find no one with one. I appreciate any help you can give me. Thank you.
I find no mention of male varieties, only female and self-fertile varieties such as Triumph. Your male plant may actually be a self fertile variety, and if so, will produce fruit without the female. These articles will help:
